What is the Cambridge Structural Database (CSD)?
The CSD (Cambridge Structural Database) is a commercial CCDC database with 1.3 million organic and metal-organic structures — the gold standard of crystallography. Every structure has a unique refcode (e.g. ACSALA01 = aspirin Form I). Access requires a licence (~USD 5,775/year academic). WebCSD (searching individual structures) and Mercury (visualisation) are free.
What is Rietveld refinement and when is it used?
Rietveld refinement (Hugo Rietveld, 1969) is a method of fitting the whole PXRD pattern to a structural model by least squares. It makes it possible to determine lattice parameters, atomic positions and the percentage phase composition of a mixture. It is used when you need more than identification — for example to quantify polymorphs or to refine a structure from powder data.
What is a CIF file and what does it contain?
CIF (Crystallographic Information File) is the standard format for crystallographic data (IUCr, 1991). It contains: unit cell parameters (_cell_length_a/b/c, _cell_angle_*), the space group (_symmetry_space_group_name_H-M), atomic coordinates (_atom_site_*), the R-factor, the publication DOI and optionally the PXRD pattern (_pd_peak_2theta). Parsing CIF is the starting point for any integration with crystallographic databases.
How are polymorphic forms of a compound distinguished by PXRD?
Different polymorphic forms have different unit cell parameters, and therefore different 2θ peak positions. Just 3–5 characteristic peaks are enough to identify a form unambiguously. For example, aspirin Form I has a strong peak at 15.52°, Form II at 10.36°. The method is used as standard in the pharmaceutical industry (USP <941>, PhEur 2.9.33).
